Research Project: Analysis of Soybean Genes Involved in Pest Resistance

Objective:
The object is to identify soybean genes and resistance mechanisms involved in the defense response of specific soybean genotypes to the soybean cyst nematode race 1 and race 14.

Approach:
Our soybean Expressed Sequence Tag (EST) database will serve as the foundation for identifying genes involved in resistance in the soybean cyst nematode. EST inserts will be amplified by the polymerase chain reaction and will be imprinted on glass slides. Fluorescently labeled probe derived from mRNA from soybean treated and not treated with soybean cyst nematodes race 1 and 14 will be hybridized to the array. The amount of expression of each gene will be quantitated. Correlations will be made among gene expression patterns and quantity in relation to gene identity, location and timing of expression, and other parameters to determine gene function and mechanisms of resistance. Molecular data will be correlated with cytological data using bright field and electron microscopy.
